Binding within the Nuclear Annuli and Its Possible Effect on Nucleocytoplasmic Exchanges
نویسنده
چکیده
Evidence exists which indicates that the pores of the nuclear envelope are important pathways for macromolecular exchanges between the nucleoplasm and cytoplasm (1-4). The pores as seen with the electron microscope often contain an electron-opaque material (5-7), and it has been suggested that the presence of such pore material could affect nucleocytoplasmic exchanges (8). Investigations in which the passage of colloidal particles through the nuclear annuli of intact amebas was studied support this hypothesis, and further suggest that the colloidal particles bind to the material within the pores (3). If such binding does occur, it could be an important factor in controlling macromolecular exchanges between the nucleus and cytoplasm. To determine the extent to which binding to the pore material can occur, experiments were conducted in which isolated nuclei or isolated nuclear envelopes were treated with protected colloidal gold particles and subsequently examined with the electron microscope.

The Effect of the Electron-opaque Pore Material on Exchanges through the Nuclear Annuli
To investigate the extent to which the electron-opaque pore material can regulate nucleocytoplasmic exchanges which occur through the nuclear annuli, experiments were performed in which polyvinylpyrrolidone (PVP)-coated colloidal gold particles (25 to 170 A in diameter) were microinjected into the cytoplasm of amebas (Amoeba proteus). The cells were fixed at various times after injection and ex...

متن کاملThe Nuclear Annuli as Pathways for Nucleocytoplasmic Exchanges
Colloidal gold particles, 25 to 55 A in diameter, which had been coated with polyvinylpyrrolidone, were microinjected into the ground cytoplasm of amebas (Chaos chaos). At time intervals of 1 minute, 2 minutes, 10 minutes, and 24 hours after injection the cells were fixed for electron microscopy. After 24 hours, gold particles were found in both the nuclei and the ground cytoplasm, the concentr...
متن کاملPossible Role of Intranuclear Membranes in Nuclear-cytoplasmic Exchange in Spider Crab Oocytes
It is well established that ribosomes and most of the mRNA needed to support protein synthesis during early stages of development are synthesized during oogenesis (Brown and Littna, 1964) . Large quantities of rRNA and its associated proteins are synthesized by the nucleolus and transferred from nucleus to the cytoplasm (Perry, 1966). How rRNA is transferred from nucleus to cytoplasm remains ob...
متن کاملSox10 is an active nucleocytoplasmic shuttle protein, and shuttling is crucial for Sox10-mediated transactivation.
Sox10 belongs to a family of transcription regulators characterized by a DNA-binding domain known as the HMG box. It plays fundamental roles in neural crest development, peripheral gliogenesis, and terminal differentiation of oligodendrocytes. In accord with its function as transcription factor, Sox10 contains two nuclear localization signals and is most frequently detected in the nucleus. In t...
